Well colonisation was very important because the US started as a colony, in fact a number of colonies, split between Britain, France and Spain (various parts changed hands over the years, but to give you an idea, here is a link to a colonial map: http://static.howstuffworks.com/gif/willow/history-of-south-america0.gif ). So basically, the US would not exist if it wasn’t for colonisation.
In addition, the US itself has colonised, for example, Hawaii, so that too is obviously an important part of the formation of the US, as in the case of Hawaii, there would be less states if they had not colonised it!
I’m not sure what you mean about “unique cultures”? I think it might be that because of immigration (both forced, e.g. slavery, and voluntary, e.g. Irish and Italian immigrants) there are a number of diverse cultures that have come together to form the US as it is today. One example would be in relation to the civil rights movement, which started because of the discrepancy between how the different ethnic groups were treated, and the question is if this would have come about if it were not for the diverse nature of the US population. In other words, the multiplicity of cultures has formed the nation of tolerance (well, tolerant in theory anyway) which exists today.
